Five techniques for preparing inocula of Meloidogyne incognita and M. hapla were compared. These were: (i) light brown intact egg masses, (ii) egg masses fragmented with a Ten-Broek tissue homogenizer, (iii) eggs collected by the Coolen & D'Herde procedure, (iv) larvae collected by incubating infected roots in a mist chamber for 8 to 12 hours and (v) eggs collected by dissolving gelatinous matrices in sodium hypochlorite solution at concentrations of 0.53%, 1.05% or 2.62% at 21 deg C.
