2. The metabolism and transport of fructose in small intestine were studied in guineapigs given fructose for up to 20 days. A preparation of villus epithelial cells was used for the measurements. Most of the fructose metabolized by guineapig small intestine appeared as glucose, the rest as lactate. Fructose feeding had no effect on the protein content of the epithelial cell preparation, but the DNA content decreased noticeably after 10 days on the fructose diet. Fructose metabolism decreased initially on fructose feeding and subsequently a gradual increase occurred which became pronounced after 10 days on the fructose diet. Fructose-1-phosphate aldolase in the small intestine. The rate of fructose uptake in segments of intestine from animals fed on fructose for 3 days was the same as that of animals on normal laboratory chow. There was a slight increase in the rate of fructose uptake in intestinal segments from animals given fructose for 20 days. Histological section of intestine taken from animals given fructose for 20 days showed accumulation of a yellowish material in the sub-epithelial region at the tips of the villi; it appeared to be present within macrophages. Preliminary evidence indicates that the material is a protein containing a carbohydrate component.
