Doubling chromosome numbers of wheat-rye amphiploids with colchicine, DMSO, and cold treatment

Amphidiploid seedlings were obtained following culture of hybrid embryos derived from Triticum durum X rye and T. aestivum X rye, involving ten tetraploid and five hexaploid wheats and various diploid ryes. The main tiller of seedlings grown in pots, when 10 cm tall, was cut back to 2 cm from the crown and a vial containing 0.1% colchicine in an 2% aqueous solution of dimethyl sulphonate was inverted over the cut stem.
