A method is described for immobilizing the beta -galactosidase from E. coli on Sepharose 4B activated with CNBr. The activities of the free and immobilized enzymes were measured using O-nitrophenyl beta -D-galactopyranoside or lactose (labelled on the glucose residue) as substrates. In the latter measurement, the hydrolysed sugars were separated by paper chromatography and estimated by liquid scintillation counting.
