A routine HPLC method is described. Fat is extracted and alkali-saponified, non-saponifiable matter is isolated, purified with digitonin-Celite and Florex-XXS column chromatography then by TLC of the benzene Florex fraction, and then vitamin D is determined by HPLC on a Zorbax SIL column. Samples from dried milks commercially fortified with vitamin D were co-chromatographed with authentic vitamin D2; a single peak resulted, confirming that vitamin D2 is used for commercial fortification.
