Abstract
Young leaves were sampled from species of Nicotiana, Petunia, Lycopersicon, Solanum and Capsicum. The subunit polypeptides of ribulose 1,5-disphosphate carboxylase, purified by microimmunodiffusion, were separated by isoelectric focussing. This method is considered suitable for the rapid screening of somatic hybrid plants for the expression and identification of nuclear and chloroplast nucleic acids. Sample size may be as small as 1 g leaf tissue. Another method for identifying the genetic constitution of organelles is described, by which purified DNA is digested with the restriction endonucleases R. Hind III or R. Eco RI, and the resultant fragments are fractionated by gel electrophoresis.
