7,8-Dihydropteroate-synthesizing enzyme from Plasmodium chabaudi

Assays of 7,8-dihydropteroate-synthesizing enzyme activity in P. chabaudi extracts are described. Dihydropteroate was synthesized by both crude and purified preparations from 2-amino-4-hydroxy-6-hydroxymethyldihydropteridine, ATP and p-aminobenzoate. Both the kinase and synthase reactions were catalyzed by a single enzyme or an enzyme complex. 2-Amino-4-hydroxy-6-carboxydihydropteridine was a potent inhibitor, Optimal activity was at pH 8.5 to 9.0.
