( alpha 1 'right arrow' 2)-, ( alpha 1 'right arrow' 4)- and ( alpha 1 'right arrow' 3)fucosyltransferases (FT) were separated from skim milk using an ion-exchange column containing sulphopropyl-Sephadex C-50 and were purified resp. 18 000-, 39 000- and 56 000-fold on columns containing GDP-hexanolamine-agarose. ( alpha 1 'right arrow' 2)-, ( alpha 1 'right arrow' 3)- and ( alpha 1 'right arrow' 4)FT had pH optima of resp. 5.9-6.2, 7.6-8 and 7.6-8; the ( alpha 1 'right arrow' 3)- and ( alpha 1 'right arrow' 4)- but not ( alpha 1 'right arrow' 2)FT required Mn2+. Apparent Km of ( alpha 1 'right arrow' 3)FT for lactose, 2'-fucosyl-lactose, lactofucopentaose II, lacto-N-tetraose and lactofucopentaose I were resp. 83, 20, 17, 5 and 1.4 mM.
