To determine the cytotoxicity of 7,12-dimethylbenznthracene (DMBA) and N-nitroso-N-methylurea (NMU) on primary cultures of rat mammary cells, cultures were exposed to various concn. of these carcinogens. Cytotoxicity was evident after exposure to 24 h to as little as 0.1 mu g DMBA/ml. Cytotoxicity was evident after exposure for 2 h with NMU at 80-160 mu g/ml. Primary cultures of rat mammary cells were also examined for their ability to metabolize MBA into water-soluble products.
