Lenacil is produced by condensation of cyclohexylurea with butyl 2-oxocyclopentanecarboxylate. Fractionation by TLC on Kieselgel F254, with CHCl3-ethyl acetate-benzene (9:6:5) as mobile phase, showed that urea (RF0.1) and cyclohexylamine (RF0.9) are the main impurities in the technical product, with lenacil at RF0.6. Quantitative analysis is achieved by comparing the absorbance (at 270 nm) of a methanolic solution containing about 2 mg/litre of lenacil with that of a standard solution.
