Microsomal vesicles isolated from maize coleptiles had an Mg-ATP-dependent proton transport mechanism which was measured by quenching of quinacrine fluorescence and 14C-methylamine accumulation. The proton pump was temp.-sensitive, had a pH opt. of 7.5, was stimulated by Cl, insensitive to k and inhibited by carbonyl cyanide-m-chlorophenylhydrazone. Mn was almost as stimulatory as Mg2+ but high levels of Ca2+ were inhibitory.
