Somaclonal variation and reaction to Verticillium wilt in Medicago sativa L. plants regenerated from protoplasts

Lucerne (alfalfa) plants, M. sativa L., regenerated from mesophyll protoplasts were of 2 morphological types; normal, resembling the parental type, and variants. Cytogenetic studies, coupled with measurements of pollen grain and guard cell dimensions established that variant protoclones were of higher ploidy than normal. Inoculation tests showed that the variant clones were highly tolerant to the wilt fungus V. albo-atrum. The enhanced tolerance is associated with an increase in ploidy and thus might result from a gene dosage effect.
