Callus formation and somatic embryogenesis from leaf cultures of orchardgrass (Dactylis glomerata L.)

The frequency of production of callus and regenerated plants declined as leaf sections were taken at increasing distances from the shoot apical meristem. Somatic embryos, which arose from mesophyll tissue, showed a higher embryogenic potential than seed-grown plants. Photoperiod and temperature significantly affected the frequency and extent of callus formation. In a study of embryogenic genotypes, initial culture temperatures did not affect embryogenesis.
