Callus induction from barley microspores

The barley anther culture (BAC1) liquid basal medium, supplemented with growth regulators and replenished at 10-day intervals was optimal for anther response and productivity. An inoculation density of at least 20 anthers/ml and culture at 28 degrees C in darkness further improved results. Under this regime, Elrose produced 591% calluses (as a percentage of anthers plated), while Klages and Bruce produced 918% and 40%, respectively, in BAC1 medium containing 20 g Ficoll/litre.
