Using an agar well assay technique (British Standards Institution (1968) BS 4285), a standardized spore suspension prepared from Bacillus stearothermophilus subsp. calidolactis C-953 was added at 2% to antibiotics medium no. 5 (Streptomycin assay agar, Hi-Media, Bombay). 12 ml of the inoculated medium was poured into standard assay plates (20 x 100 mm) and wells of 8 mm diam. were bored in the solidified agar.
