Isolation and purification of soybean leaf urease
Soyabean leaf urease was extracted by pulverizing the leaves in liquid N2 and extracting with 1 vol. buffer. The crude extract was centrifuged, heated (65 degrees C in 15 min in a microwave oven) and fractionated on QAE25 Sephadex. The enzyme was purified >500-fold. Compared with the seed enzyme, the leaf enzyme showed differences in electrophoresis, inhibitor sensitivity and kinetics.
