The method of staining pollen nuclei

Current Science 53(13): 704-705

The method entails placing pollen grains (either freshly collected or previously fixed in Carnoy solution) in a drop of lactoproprionic orcein on a slide, covering with a glass cover slip and leaving for 24 h before examination under a light microscope. The protoplast of the pollen grain passes, with unruptured intine, through the pollen pore and separates from the exine; the nuclei stain intensely.
