Attempted cryopreservation of Leishmania major amastigotes in the ear skin of experimentally infected golden hamsters (Mesocricetus auratus)

Ears of golden hamsters with marked leishmanial lesions were either injected with cryopreservant (phosphate buffer pH 7.8 with 10% glycerin) and then immersed in preservant for 10 or 60 min, or immersed in the preservant for 10 or 60 min before freezing at -170 degrees C in liquid nitrogen vapour or frozen without preservant. Survival of L. major amastigotes was assessed one month and one year later on parasite growth in NNN medium and on the presence of unchanged amastigotes in tissue prints.
