Quantitative determination of primary amino groups in amino acids, peptides and proteins is made by conversion using o-phthaldialdehyde in the presence of a thiol component into bisalkylated isoindoles (OPA method). Since mercaptoethanol, normally used as thiol reagent, often causes unstable extinction values, it was substituted by ethanthiol and the extinction coeff. of 20 amino acids determined.
