A semi-automated continuous flow method for the determination of the activity of alkaline phosphatase in soils is described. The soil is buffered to pH 10.0, phenolphosphate disodium salt is added as a substrate and the batch is incubated by using a rotating shaker in an incubator at 37 degrees C for 5 hours.
