Cytoplasmic protein phosphorylation and prolactin (PRL) synthesis and release were studied in rat normal and malignant pitiutary cells in response to various pharmacological agents. In tumour cells, the actions of thyrotropin-releasing hormone (TRH), vasoactive intestinal protein and EGTA suggested that phosphorylation of one set of cytoplasmic proteins (MW 60 000-80 000) was related to PRL synthesis, and phosphorylation of a second set (MW 18 000-20 000) to PRL release.
