The method of sterilizing explants from orthotropous suckers is described. Treatment of the suckers with 2% orthodifolatan before collection resulted in 95% of the cultured explants remaining healthy. There was a minimum explant size below which buds did not burst in the absence of growth regulators in the culture medium. No callus formed on basal medium unless zeatin was present; zeatin also promoted bud elongation. IAA resulted in little leaf development and in rapid leaf abscission.
