The techniques used for the culture of hypocotyl-derived callus cells and the isolation, culture and fusion of protoplasts from them are described. Of the 25 cultivars used, Akage had the best callus growth, producing 492 x 104 protoplasts/g callus fresh weight. Only Akage showed protoplast cell division (after 13 days) and after 4 weeks produced several small colonies.
