Milk and dried milk, buttermilk and buttermilk powder, whey and whey powder. Detection of phosphatase activity (Screening method)

Milk and dried milk, buttermilk and buttermilk powder, whey and whey powder. Detection of phosphatase activity (Screening method)

International IDF Standard (82A: 1987 (Provisional)): 3

The product is diluted with a buffer substrate containing disodium p-nitrophenyl phosphate at pH 10.2 and incubated for 2 h at 37 degrees C. Any p-nitrophenol liberated by alkaline phosphatase is then detected by its yellow colour, either visually or spectrometrically. Unlike the reference method (IDF Standard 63:1981), this method is not affected by phenol.
