Following a summary of the history of sister chromatid exchange (SCE) research, some of the problems approached using the 5-bromodeoxyuridine (BrdUrd) technique are reviewed under the headings (1) the mechanism of sister-chromatid differentiation, (2) BrdUrd incorporation into plant DNA and its effects on cell cycle kinetics, (3) chromosome replication banding, (4) the frequency of SCEs in different plant species, (5) SCEs: spontaneous or induced? (6) 'dot-like' SCEs and chromosome structure, (7) SCE distribution along chromosomes, (8) factors affecting the frequency of SCEs in higher plant cells, (9) the SCE test in cytogenetic toxicology, (10) sister-chromatid differentiation and SCEs related to the meiotic process and (11) the mechanism of SCE formation.
