Tissue culture of sugarbeet. Effects of culture temperature, pH, sucrose content, sugar and herbicide on callus growth

In experiments using 4 callus culture lines on Murashige & Skoog medium with added IAA and benzyladenine, the optimum culture temperature ranged from 25 degrees C to 30 degrees C. The best pH of the medium was about 6 and the best sucrose content about 2%. Sucrose was the best carbon source among 10 sugars examined. Callus growth was inhibited by 0.5 mM glyphosate added to the culture medium.
