A correlation between the site of Tn5 insertion in a 14 kb fragment from the Sym plasmid of strain ANU843 and the resulting nodulation phenotype led to the identification of 3 regions on the plasmid, designated I, II and III, which affected nodulation. Region I mutants lost normal root hair curling ability and were unable to induce infection threads, so making them unable to nodulate any plant tested (clovers and pea).
