Physiological effects of callus tissues on the subcultures: polyphenols and enzymes

From the sweet cherry cultivar Sam, a segment 0.5-mm thick from a 1-year-old shoot was taken, the phloem and outer tissue removed and the remainder sterilized and grown in vitro; 15 mg/litre of indole acetic acid (IAA), 0.2 mg/litre of benzyladenine and 5, 10 and 30 mg/litre of ACC were applied. After being kept in the dark for 4 weeks, the tissue was separated into older and younger segments of callus and subcultured for 4 weeks in the dark.
