The M5 RNA of bluetongue-virus-2 (BTV-2) was deduced from sequence analyses to be 1635 base pairs long, three nucleotides shorter than the M5 gene of BTV-10 U.S. but the same as serotype 1 (South Africa and Australia). The amino acid sequence encoded by the cDNA yielded a protein of 526 amino acids of molecular size 58 954 daltons and short non-coding regions. The charge of the protein is +1.5 at neutral pH compared with -4.5 of BTV-10. The amino acid sequence comparisons of BTV-1 SAfr. and Aus, BTV-2 and BTV-10 used the BTV-10 sequence as a reference. The homology between BTV-1 Aus and BTV-2 U.S. is up to 96%; both molecules contain three cysteine residues at the same positions. Similarly the variability in VP5 between BTV-1 SAfr. and BTV-2 U.S. is only 10%.
