Differential sensitivity of stomata and mesophyll to sudden exposure of bean shoots to ozone

Shoots of single intact dwarf bean plants (Phaseolus vulgaris L.) were exposed to O3 concentrations 0.16 - 0.64 cm3 m-3 for 3 - 4 h under limiting or saturating CO2 concentrations. Water vapour (E), CO2 (F) and O3 exchange rates of the shoots were monitored during the exposures. After the treatment changes in the water relations of stomatal and mesophyll cells were extimated by measuring the responses of E and water content of single leaflets to hydraulic pressurization via xylem. E and F of the shoots were converted to stomatal (gs) and mesophyll (gm) conductances averaged over all leaves, using a sophisticated flow-resistance approach. Under O3, gs decreased precipitously, with some transient recovery before the final decline. The decline was more pronouced under high CO2 and O3 concentrations, and highly correlated with the totals of O3 absorbed, and more loosely with the above-leaf concentration totals. gm was not reduced to the highest totals of O3 absorbed, although the swelling capcity of the mesophyll cells was impaired. Thus the closing of the stomata under O3 was not mediated by a dysfunction of the mesophyll photosynthesis but caused by the absorption of O3 on the wat surfaces of the stomatal cells where toxic derivatives of this oxidant caused changes in cell wall properties.
