Effect of granulosa cells added to culture medium for in vitro maturation and fertilization of bovine oocytes

As factors for better in vitro fertilization rate, effects of the presence of cumulus cells, and the number of granulosa cells or oocytes added to culture medium, were investigated together with changes in progesterone (P4) and estradiol-17.beta. (E2) concentrations in the medium. Fertilization rates of cumulus-denuded oocytes were significantly (P < 0.05) lower than cumulus enclosed oocytes (27.5% vs 55.0%). The increased numbers of oocytes per culture well significantly (P < 0.05) lowered fertilization rate. Oocytes cultured in the medium supplemented with granulosa cells had significantly (P < 0.05) higher rates of fertilization compared with those cultured without the cells. The concentrations of P4 and E2 increased as increasing densities of granulosa cells. P4 concentration was not changed with varied numbers of oocytes, but E2 concentration was highest in the absence of oocytes. The present results indicate that cumulus cells, numbers of cultured oocytes in maturation medium, and the addition of granulosa cells to culture medium are factors affecting both nuclear and cytoplasmic maturation of bovine oocytes.
