A new method to determine leiocarposide content in S. virgaurea based on the Emerson reaction is described. Leiocarposide, when reacted with aminopyrazolone and an oxidizer in alkaline conditions, gives a product stable in chloroform with a UV absorption maximum at 455 nm. There was a close linear relationship between leiocarposide concentration (C, in the range 0.002-0.020 mg/ml) and the extinction coefficient (E), viz. C = 0.0414573 E.
