Study on the regulation of potato tuber pyrophosphate: fructose 6-phosphate-1-phosphotransferase by fructose 2,6-Bisphosphate and other compounds

Pyrophosphate-fructose-6-phosphate-1-phosphotransferase (PFP) was purified from potato tubers by various methods. Fructose 2,6-bisphosphate (FrU2,6P2) strongly activated PFP. Vmax of PFP increased 2.5-fold in the presence of 1 micro mol Fru2,6P2/litre while Km of PFP for inorganic pyrophosphate decreased 6.1-fold and S0.5 for fructose-6-phosphate decreased 4.5-fold. Mg2+ activated PFP at low concn but inhibited it at high concn (>0.2 mmol/litre). Ca2+ was a competitive inhibitor with Mg2+.
