Targeted insertion of the neomycin phosphotransferase gene into the tubulin gene cluster of Trypanosoma brucei

Targeted insertion of the neomycin phosphotransferase gene into the tubulin gene cluster of Trypanosoma brucei

Nature London 348(6297): 174-175

Two gene-targeting plasmids, pUCTbneo2 and pUCTbneo3, were constructed which contained an in-frame fusion of the T. brucei tubulin gene and the Escherichia coli neomycin resistance gene (neor) gene. The plasmids were introduced into procyclic trypanosomes by electroporation, either as supercoiled DNA or after linearisation. Targeting efficiency was higher with a linearised than with a circular construct, and with 5 kb of tubulin gene cluster than with 0.9 kb.
