A successful transfer of exogenous DNA into chickens by microinjection into the germinal disc of fertilized ovum

Plasmid DNA containing the E. coli beta -galactosidase (lacZ) gene under the control of fowl beta -actin gene promoter was injected in linearized from into the germinal disc of fertilized fowl ova at the single-cell stage, obtained from the magnum. The embryos were cultured in vitro and then in surrogate eggshells to hatching. The rate of hatching was 11.08% (31/263); 19 mature birds were obtained.
