Generation of transgenic dairy cattle from transgene-analysed and sexed embryos produced in vitro

Bovine oocytes were isolated from slaughterhouse-derived ovaries and were matured and fertilized in vitro. They were then microinjected with a dam-methylated gene construct consisting of genomic sequences encoding human erythropoietin, under the control of bovine alpha S1-casein regulatory sequences. After 6-7 days in culture, the embryos were biopsied for sexing and transgene analysis. The transgene analysis was accomplished by treatment of embryo lysates with DpnI and Bal31, followed by PCR.
