Genetic analysis of Candida albicans strains studied by isoenzyme electrophoresis

The polymorphism of homologous proteins (isoenzymes) presented by 21 loci is analysed for the characterization of the genetic structure of Candida albicans natural populations. Fifty five C. albicans strains isolated from oral thrush of 55 HIV+ patients are characterized. Among the 21 genetically interpreted loci, 13 are polymorphic, which made it possible to differenciate 41 genotypes of "zymodemes". Amogn the 55 strains, 39 display one specific genotype, while the 16 remaining strains are found to belong to two other genotypes: the GRI genotype was found in 13 strains (24% of the strains tested); the GR2 genotype was found in 3 strains (5%). The genetic characterization of C. albicans strains is 100% reproducible. Moreover, no genotype modification is observed during the laboratory maintenance of strains. This methodology showed strong genetic variability between the strains studied. This variability analysed by a factorial correspondence analysis (AFC) revealed a definite population structuring. The isoenzyme polymorphism analysis is thus an efficient and reproducible method. It leads to the characterization of different C. albicans strains with a great discriminatory power. This method proved to give relevant genetic informations, allowing a better knowledge of the biology of this species.
