Genetic engineering method for diagnosing fertile and sterile cytoplasm of Helianthus annuus at early stages of ontogeny

Mitochondria of fertile plants differ from those of their cytoplasmically male-sterile counterparts with cytoplasm of Helianthus petiolaris in having plasmid-like DNA (the PlT-plasmid). In studies using A, B and C lines, the PlT-plasmid was identified only for plants of 8 lines. It was cloned for the BamHI site of plasmid Puc18 and the recombinant plasmid was used to construct a probe which allowed reliable identification of material containing the plasmid mitochondrial DNA of H. annuus.
