Genetic linkage of the Rps1 and L2 loci in soybean

Genome mapping of important genes in soybean, Glycine max (L.) Merr., is rather limited compared with that of other major crops. Such genetic information is needed by those involved in soybean genetic improvement. In this study, genetic linkage of the Rps1 locus controlling reaction to Phytophthora megasperma Drechs. f. sp. glycinea T. Kuan and D.C. Erwin, and the L2 locus controlling pod wall color was tested. Populations segregating for both genes were derived from the cross Bedford times D78-3173. At maturity the pod wall color of F-2 plants was recorded. The F-3 seedlings derived from those plants were evaluated in a greenhouse for reaction to race 1 of P. megasperma using the hypocotyl puncture inoculation method. The chi-square test for linkage was significant (P lt 0.0005). Recombination was calculated by the ratio of products methods (27 +- 4.3%) and maximum likelihood method (28 +- 4.1%), indicating that these genes are on the same chromosome.
