Aureobasidium pullulans LV10 produces extracellular amylolytic activity in the fermentation liquid when grown on medium containing 3% (w/v) cassava flour as the sole carbon source. Maximum enzyme activity (1.6 AU/ml) was obtained after 40 h of static culture at 26 degree C. The enzyme was a glucoamylase which showed a preference for amylose and soluble starch but not hydrolyzed beta cyclodextrin; glucose was the only low molecular weight product of the enzyme action. The enzyme preparation showed optimum catalytic activity at pH 5.2 and at 45 degree C. Enzyme activity was increased by addition of Ca++ whilst was inhibited by Hg++. The enzymic preparation and fungal biomass, free or immobilized in calcium alginate gel beads, were used for the hydrolysis of raw cassava flour. Moreover, by our fermentation process, cassava flour was transformed into a product (biomass) containing 28-30% of crude protein with a 48% conversion yield. All essential amino acids were present in the biomass which had a biological value of 82%.
