Investigation of the medium for barley mature embryo culture
In mature embryo cultures of 13 barley cultivars, frequency of callus production varied with cultivar and with media composition. Factors affecting callus production included the alanine and potassium dihydrogen phosphate concentrations and the type and level of growth substances. The effectiveness of growth substances varied with the basal medium used. Rapid callus growth often gave low quality callus. High callus induction frequency was not always associated with high regenerative ability.
