An adaptation of the modified dry blot technique for RFLP analysis of maize DNA is described. Starting from DNA digestion, the amount of time to detect single-copy sequences with 32P-dCTP nick translocated probes is reduced to a minimum of 4 days. Gene Screen Plus membranes can be stripped and rehybridized 8 or more times, maintaining signal intensity without increasing background.
