Studies on the targeted mutation of the transforming growth factor beta 1 gene in embryonic stem cells, followed by the breeding of mutant mice, are reviewed. Homozygous mutant mice had multifocal inflammatory disease, caused by abrogation of the anti-inflammatory and immunosuppressive effects of the cytokine.
