A new method for determining ploidy levels in cytochimaeral plants was developed based on the maximum number of nucleoli per mesophyll cell. Colchicine-treated plants of Allium wakegi and A. fistulosum, which have different ploidy levels in the first (LI), second (LII), and third (LIII) layers of the apical growing point, were used together with untreated 2-2-2 plants of the same species, which were diploid in each of these three layers. Nucleoli in guard cells of the third youngest leaf for LI and in mesophyll cells for LII were stained in a 50% AgNO3 aqueous solution at 55 degrees C for 3 h under dark, humid conditions.
