Effect of sperm quality on the efficiency of sperm separation using BSA gradient

X and Y chromosome-bearing sperm populations of cattle were separated using the active migration into a bovine serum albumin (BSA) gradient. The results of separation demonstrate that the quality of sperm especially sperm mobility influence the efficiency of separation. Both the high individual variability of separation success and the high correlation (r = 0.92) between the enrichment rate of Y-bearing spermatozoa and the calculated sperm quality index indicate a significant influence of cellular factors (maturity or age of sperm) on sperm separation using BSA gradient. A preselection of viable and most motile spermatozoa using swim-up technique resulted both a significant increase of the proportion of Y chromosome-bearing spermatozoa and a significant reduction of the correlation coefficient (r = 0.11) between the enrichment rate of Y-bearing spermatozoa and the calculated sperm quality index.
