Effect of storage on the physical and biochemical parameters of broiler semen

Pooled semen samples from 25 synthetic broiler cocks were examined for their physical and biochemical quality after storing for 0, 6, 12, 24 and 48 hr at 4-6 degree C. Initial motility of spermatozoa was drastically reduced whereas the live abnormal count increased drastically. There was a significant decrease in the percentage of live spermatozoa. The methylene blue reduction time markedly increased. Electrophoresis of seminal plasma protein revealed the presence of an additional new protein band in all the stored samples. There was a moderate but significant increase in seminal plasma acid phosphatase activity during first 6 hr of storage after which there was slight fall in activity, but the alkaline phosphatase activity remained constant. There was moderate rise in inorganic phosphorous and fall in magnesium concentration of seminal plasma during storage, but the calcium content could not be detected. These results suggest a possibility that the undiluted semen can be stored at 4-6 degree C up to 6 hr without significant deterioration in quality.
