In-vitro shoot multiplication from cotyledonary node explants of Tecomella undulata

A method is described for in-vitro micropropagation through cotyledonary nodes of Tecomella undulata, an ornamental arid tree valued for high quality timber. Various explants were excised from 6-8 days old in-vitro grown seedlings raised from superior seed stocks. Shoot bud induction from cotyledonary node explants was observed on Murashige and Skoog (MS) medium containing various treatments of growth regulators. A maximum of 26 shoots was obtained on MS medium supplemented with 3-indole acetic acid (0.05 mg/1) and benzyladenine (2.5 mg/l). Multiplication and elongation of shoots were achieved when transferred to MS medium containing low level of BA (1.0-1.5 mg/l). Well developed leafy shoots were rooted on growth regulator-free MS medium (half strength) after an exposure of auxin mixture (IAA, IBA, and NAA, 5.0 mg/l, each) for 24-48 hrs without callus formation. An increase in treatment period or amount of auxin promoted callussing of shoots. Maximum 76% rooting was obtained after 3 weeks of pretreatment. In-vitro developed plantlets were transferred to field conditions.
