Plant regeneration from petiole protoplasts of sweet potato (Ipomoea batatas (L.) Lam.) and related species

Protoplasts from petioles of sweet potatoes Kokei 14, I. triloba and I. lacunosa were cultured on MS medium with 0.05 mg 2,4-D and 0.5 mg kinetin/litre. A high frequency of callus formation was obtained and at 8-12 weeks after plating calluses 2-3 mm in diameter were transferred to MS medium with 0.2 mg IAA and 1 or 3 mg BA/litre for 3-6 weeks. Plants were produced by 14-18% of I. triloba cultures.
