A microinjection system for the analysis of gene expression in single barley coleoptile cell

Among glass pipettes prepared with a needle-drawing apparatus, those with tip diameter of 2.5-3 micro m were most suitable for sucking up cytoplasmic contents of barley coleoptile cells. The volume of sucked up cytoplasmic contents was proportional to the number of microsucked cells, and the volume of cytoplasmic content of a single cell was estimated to be approximately 2.8 x 10-2 nl.
