In sheep, an electrophoretic variant at the alpha s1-casein locus, designated CASA1 (Welsh), has been reported previously, and later studies have shown that the Welsh variant is due to a Ser-68->Asn-68 substitution. To develop a DNA-based method to identify carriers of the Welsh variant, allele-specific primers were designed to include the part of the 9th exon of the CASA1 gene that is responsible for the Ser-68->Asn-68 substitution.
